冒険・友情といったテーマを前面に掲げ、少年漫画の王道を行く物語でありながらも深く練り込まれたストーリー設定であり、随所に感動エピソードやバトルシーン、ギャグなどを織り交ぜた少年漫画として人気を博している。

2010年1月現在、原作の単行本は56巻まで刊行されており、『ジャンプ』現行連載陣の中では『こちら葛飾区亀有公園前派出所』（1976年から連載中）に次ぐ長期連載となっている。単行本売り上げは、史上最速1億部突破を達成し^[1]、初版発行部数285万部【56巻】と累計発行部数1億7900万部以上【56巻時点】の日本記録を保持している^[2][3]。

1999年に放送が始まったアニメ版もロングラン作品になっている。2007年には、連載10周年を記念して5冊の本が出版される等、様々な企画が立てられた。 

A boy named Monkey D. Luffy, inspired by his childhood hero "Red-Haired" Shanks, sets out on a journey to find One Piece, the legendary treasure of the King of the Pirates, Gol D. Roger. To accomplish this, he must reach the end of the most deadly and dangerous ocean: The Grand Line.

Luffy captains the Straw Hat Pirates first through the sea of East Blue and then through the Grand Line. He follows the path of the deceased King of the Pirates, Gol D. Roger, from island to island on his way to the great treasure One Piece. On his way his crew grows to have a firstmate - swordsman "Pirate Hunter" Roronoa Zoro, a navigator - "Cat Thief" Nami, a sniper-"Sogeking" Usopp, a cook- "Black Leg" Sanji, a doctor- "Cotton Candy Lover" Tony Tony Chopper, an archaeologist- "Demon Child" Nico Robin, a shipwright- "Cyborg" Franky, and a musician- "Dead Bones" Brook.

During the course of the story, the crew contends with both other less moral pirate crews and the Marines. The latter are the subordinates of the corrupt World Government, who apparently seek justice by ending the Golden Age of Pirates. Many background story elements involve the delicate balance of power between the World Government and the world's most powerful pirate crews, especially the Yonkou, the four most powerful pirates in the world.
